Article Snippet: For differentiation into M0 macrophages, cells in the logarithmic growth phase were seeded at a density of 1 × 10 6 cells per well in 6-well plates (approximately, 1 × 10 5 cells/cm 2 ) and treated with 100 ng/mL phorbol 12-myristate 13-acetate (PMA, HY-18738, MCE, USA) for 48 h; adherent cells were then gently washed twice with PBS and incubated in fresh complete medium for an additional 24 h. M1 polarization was achieved by stimulating M0 macrophages with 100 ng/mL lipopolysaccharide (LPS, HY-D1056, MCE, USA) and 20 ng/mL interferon-gamma (IFN-γ, HY-P7025, MCE, USA) for 48 h.
